Repair Cracking Grout

I have a home that is about two years old and in the master shower I have stone tile.

I have noticed that the grout is cracking and some of the tile is starting to separate from the wall on the outer edges of the shower. Can I re-grout and be O.K?  I am afraid that I will be getting moisture behind the tile. I know that the builder did not caulk the bottom of the shower glass door, so now I also have a problem of some mold on the outside. What can I do?
